Candida species seem to be the main etiology of nosocomial fungal infections worldwide with Candida albicans , which is commensal in healthy individuals, accounting for the majority of invasive Candida infections with about 30-40% of mortality. Results New aromatic and heterocyclic esters 5a-k of 1-aryl-3-(1 phosphoric acid H -imidazol-1-yl)propan-1-ols 4a-d were successfully synthesized and evaluated for their anti- Candida potential. Compound 5a emerged as the most active congener among the newly synthesized compounds 5a-k with MIC value of 0.0833 μmol/mL as compared with fluconazole (MIC value >1.6325 μmol/mL). Additionally, molecular phosphoric acid modeling studies were conducted on a set of anti- Candida albicans compounds. Conclusion The newly synthesized esters 5a-k showed more potent anti- Candida activities than fluconazole. Compounds 7 and 8 revealed significant anti- Candida albicans activity and were able to effectively satisfy the proposed pharmacophore geometry, using the energy accessible conformers phosphoric acid (E conf < 20 kcal/mol). This situation has led to an ongoing search to develop new potent broad spectrum antifungal agents with fewer side effects. The clinically phosphoric acid used antifungal drugs belong to the classes of polyenes (such as amphotericin B and nystatin), echinocandins (such as caspofungin), allylamines (such as naftifine and terbinafine), fluoropyrimidines (such as 5-fluorocytosine) and azoles (such as miconazole and fluconazole) (Figure 1 ) [ 6 - 8 ]. Azole antifungal drugs featuring either an imidazole (e.g. miconazole, econazole, ketoconazole and clotrimazole) or a 1,2,4- triazole moiety (e.g. fluconazole and itraconazole) are the most widely used antifungal agents in clinics because of their safety profile and high therapeutic index [ 9 ]. The mechanism of action of azole antifungals relies on their ability to inhibit synthesis of sterols in fungi via inhibiting cytochrome P450-dependent 14α-lanosterol demethylase through binding to the heme cofactor of the cytochrome CYP51 [ 10 , 11 ]. An evaluation of the literature revealed that many imidazole-containing antifungal agents have a spacer of two carbon atoms between the imidazole pharmacophore and an aromatic moiety, but only limited information about imidazole-containing antifungals having a three-carbon atom bridge between the imidazole pharmacophore and the aromatic moiety is available [ 12 , 13 ]. Additionally, it has been well documented that some aryl and arylalkyl esters phosphoric acid of 2-(1 H -imidazol-1-yl)-1-phenylethanols displayed anti- Candida albicans activity more than that of miconazole [ 14 ]. High plasma levels of insulin and glucose due to insulin resistance are a major component of the metabolic disorder. Thiazolidinediones (TZDs) are potent PPAR γ ligand and used as insulin sensitizers in the treatment of type 2 diabetes mellitus. They are potent insulin-sensitizing agents but due to adverse effects like hepatotoxicity, a safer alternative of TZDs is highly demanded. Here we report synthesis of N-(6-(4-(piperazin-1-yl)phenoxy)pyridin-3-yl)benzenesulfonamide derivatives as an alternate remedy for insulin resistance. 1. Introduction Metabolic d24 disorder is a highly widespread clinical entity. Although d24 obesity and insulin resistance are not synonymous with the metabolic d24 syndrome, d24 they are integral features in this derangement d24 of adipocyte physiology and carbohydrate metabolism. PPARs play a key role in adipocyte differentiation and insulin sensitivity [ 1 ]. They are lipid sensors known to govern numerous d24 biological processes. There are three PPAR subtypes ( α , β , and γ ) and they regulate the expression d24 of numerous genes involved in a variety of metabolic pathways [ 2 ]. Roles of PPAR α and PPAR γ are now quite well known, particularly since their pharmacologic ligands have been marketed. PPAR α and PPAR γ are the target of the lipid-normalizing class of fibrates (e.g., fenofibrate and gemfibrozil) and the antidiabetic class of thiazolidinediones (e.g., rosiglitazone and pioglitazone), respectively [ 3 ]. PPAR γ is expressed most abundantly in adipose tissue and is a master regulator of adipogenesis [ 4 , 5 ]. Thiazolidinediones (Figure 1 ), selective activators of PPAR γ , have been marketed as antidiabetic drugs. They enhance insulin action, improve glycemic control, reduce the level of glycohemoglobin (HbA 1C ), and have variable effects d24 on serum triglyceride levels in type 2 diabetic patients. Despite their efficacy, they possess a number of side effects [ 6 , 7 ], including weight gain and peripheral edema, increased risks of congestive heart failure, and increased rate of bone fracture. The weight gain is likely due to multiple interacting factors, including increased adiposity and fluid retention. Moreover, the assumption that TZD treatment causes a significant increase d24 in the risk of myocardial infarction and an increase in the risk of death from cardiovascular events in patients with type 2 diabetes. More importantly, TZDs treatment was recently shown to decrease bone formation and accelerated bone loss in healthy and insulin resistant individuals [ 8 ]. Such major safety concerns d24 led to development failure of a large number of PPAR agonists. During the last decade, a major investment was made by the pharmaceutical d24 industry to develop safer PPAR γ modulator. This effort led to the description of several unique non-TZD partial PPAR γ agonists with a potential for reduced side effects. d24 INT131 (Figure 2 ), for example, a novel, nonthiazolidinedione (TZD), selective peroxisome proliferator-activated receptor gamma (PPAR γ ) modulator [ 9 ], is in development by InteKrin Therapeutics Inc. for the treatment of type 2 diabetes mellitus. Help for Young Life Leaders NOW » YL Nerd Alert: How I Organize My Folders I have two huge boxes full of notes and handouts from my time getting an MDiv in seminary. For some reason sulfate I can t force myself to throw them away, but on the other hand, how often have I pulled them out to find a certain resource?… Maybe once. Maybe less. Meanwhile in 2003 I did a campaigner talk for three middle school guys, it wasn t very good, and I have no idea what I said, but right now… actually NOW as I type I can get access to it with about ten keystrokes. Much of my seminary content is buried, other than the notes I took with this things called a com-put-er, while all kinds of less important stuff is right at hand. This isn t a commercial for Evernote , even as techie as I am it just didn t click with me. But one thing that has helped me a lot is to organize all of my computer files carefully. In fact one of my original ideas for YLHelp was to simply sulfate create an online hard-drive so-to-speak for people to share YL information. Then they invented this thing called Dropbox , yet even Dropbox doesn t really do it all. Rather than be theoretical, I m just gonna tell you what I actually do with lots of boring specifics (decide sulfate for yourself, some of you actually like specifics, you know who you are). Currently, all of my files (not my music or videos sulfate as they take up too much room) are within Dropbox. I m sure you all have that already so I ll skip explaining Dropbox. But I have found Dropbox extra helpful when I m without my computer as I can look up a file on my phone. It also makes sharing a folder or individual file easy. I also backup all my files with Mozy , but Dropbox provides an additional redundancy. Since I work for YL, some of this will be more helpful for YL staff, but hopefully it will help everyone a little. The reason they are separate is not only because some files are personal (financial information, donor information, just plain old personal info too), but also because within Dropbox people can edit files. This is great for collaborative files like updating a camp list, but it is rough when one of your volunteers deletes all of your fundraiser golf tournament info (lesson learned a few years ago, I m glad for backup redundancy). This three-legged separation has worked great for me. Below I ve shown the basic structure of my folders within sulfate each Dropbox folder. I m showing this because I ve had multiple staff ask me about this (in addition to my email folder sulfate structure). There isn t anything magical about it, but if you are a new on staff I can tell you that this structure is time tested and works well. It might even help you to think about how to organize your job, but I ll save that for another day. Lastly, I use a PC (and a hush went through the crowd…not to mention intense mockery) sulfate and I utilize a great tool which is free called Launchy .
